Western blot was performed using Anti-CNTNAP1 Polyclonal Antibody (Product # PA5-42737) and a 160kDa band corresponding to CNTNAP1 was observed in all cell and tissue lysates tested except Mouse Skeletal Muscle, Rat Skeletal Muscle, Mouse Liver and Rat Liver [DOI: 10.1016/s0896-6273(00)81049-1]. Whole cell extracts (30 µg lysate) of SH-SY5Y (Lane 1), Mouse Brain (Lane 2), Mouse Cerebellum (Lane 3), Rat Cerebellum (Lane 4), Mouse Skeletal Muscle (Lane 5), Rat Skeletal Muscle (Lane 6), Mouse Liver (Lane 7) and Rat Liver (Lane 8) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). An uncharacterized band (*)was observed at ~40kDa in some negative tissue models.
